چکیده

The recent paper by Huang and Davisson (J. Bacteriol., 76, 495, 1958) on the distribution of lysine racemase in bacteria reports its occurrence in only a few species of Escherichia and Proteus. Various other bacteria including 22 strains of various genera of the Pseudomonadaceae did not possess such activity. I wish to report evidence for the occurrence of lysine racemase activity in a Pseudomonas sp., which produced the fluorescent pigment typical of many such isolates from soil enrichment cultures. This work was done at the University of California at Los Angeles in 1953 and 1954 but has not been reported previously because circumstances prohibited the completion of the larger planned study on the metabolism of lysine. The specific point which would seem to explain the occurrence of the enzyme in the strain utilized is that it was isolated, by enrichment techniques, for its ability to grow on DL-lysine

Distribution of lysine racemase in bacteria.

We have reported elsewhere the racemization of lysine by cell preparations of Proteus vulgaris strain ATCC 4669 (Huang et al., 1958). The simplest explanation of the results presented is that Proteus vulgaris contains a lysine racemase. The distribution of this activity in bacteria has now been investigated. Almost a hundred species, from eleven families, have been examined. Only nine species, ...

Efficient Production of Enantiopure D-Lysine from L-Lysine by a Two-Enzyme Cascade System

The microbial production of D-lysine has been of great interest as a medicinal raw material. Here, a two-step process for D-lysine production from L-lysine by the successive microbial racemization and asymmetric degradation with lysine racemase and decarboxylase was developed. The whole-cell activities of engineered Escherichia coli expressing racemases from the strains Proteus mirabilis (LYR) ...

Hydrolysis and transpeptidation of lysine peptides by trypsin.

The study of the tryptic digestion of poly-L-lysine (Katchalski, 1951; Waley & Watson, 1953) has shown that lysine oligopeptides appear duringthe reaction. A better understanding of the mode of action of trypsin on the high-molecular-weight peptide might be gained by a study of the action of the enzyme on a homologous series of lysine peptides (for a preliminary note see Katchalski, Berger & Le...

Determination of the Very High Barriers to Conformational Processes by the Neat Racemization Technique. Part 1: Barriers to Ring Inversion in Dibenzo [a,c] Dinaphtho [2,3-e; 2,3-g] Cyclooctatetraene and Hindered Rotation in 2,2'-Dimethyl Binaphthyl

A method is described by which it is possible to determine the high barrier to ring inversion and hindered rotation by neat racemization of optically active compounds when the racemization is caused by ring inversion or hindered rotation. The method is based preparation of sufficiently pure enantiomers, mainly by chromatography on swollen microcrystalline triacetylcellulose (TAC). By this t...

Synthesis of e-Aspartyl-lysines and of isoHexylamides of Aspartic Acid

Lockhart & Abraham (1956) found that two compounds which appeared to be aspartyl-lysine and aspartyl(isoleucyl)-lysine respectively were formed when bacitracin A was hydrolysed with 11 Nhydrochloric acid at 800. The aspartic acid residue in these compounds had the L-configuration and was linked to the e-amino group of L-lysine. However, both compounds carried a net positive charge at pH 7 and t...
